Η ιστιοκυττάρωση από κύτταρα με λειτουργικούς χαρακτήρες κυττάρων Langerhans, είναι κατά βάσιν πολυσυστηματική, νεοπλασματική νόσος. Η ακριβής διάγνωση πιστοποιείται με ιστολογική, ανοσοϊστοχημική, υπερμικροσκοπική και ανοσοϋπερμικροσκοπική μελέτη των δερματικών βλαβών. Λαμβάνοντας υπόψιν αφενός το δεδομένο ότι πρόκειται για πολύ σπάνια νόσο και αφετέρου τη μεγάλη ποικιλία της κλινικής έκφρασης, γίνεται κατανοητή η εξατομίκευση του βιολογικού και παρακλινικού ελέγχου. Οι μονοσυστηματικές μορφές (δερματική, οστική) χαρακτηρίζονται από καλύτερη πρόγνωση και ευχερέστερη θεραπευτική αντιμετώπιση. Οι πολυσυστηματικές μορφές έχουν σοβαρή κλινικοεργαστηριακή εικόνα και απαιτούν συνδυασμένη χορήγηση μειζόνων φαρμάκων. Η προσβολή ζωτικών για την επιβίωση οργάνων και η επέλευση ενδονοσοκομειακών - ιατρογενών (λόγω της εφαρμογής συγκεκριμένων θεραπευτικών πρωτοκόλλων) παρεμπιπτουσών λοιμώξεων, αποτελούν τους κυριώτερους παράγοντες που μπορεί να απειλήσουν τη ζωή του ασθενούς.
