EVALUATION OF THE SUBACUTE TOXICITY OF CLASSIC ANTINEOPLASMATIC COMPOUNDS IN RATTUS NORVEGICUS RATS
Abstract
One of the most attractive aspects in the use of chemotherapy for the control of malignancy is the ability to minimize side effects and to preserve an intack host.
Several attempts have been made in the past for the early diagnosis and prevention of acute and chronic side effects following cancer chemotherapy. The present thesis proposes a novel biochemical screening method for eνaluating subacute toxicity during cancer chemotherapy. Toxicological evaluation was performed in serum samples at time intervals up to 15 days, following administration of dose regimens that were previously shown to be effective in suppressing malignant tumor growth or to prolong survival in tumor bearing animals. Cardiac, hepatic and renal subacute toxicity was evaluated by measuring enzyme activity or concentration of: alanine aminotransferase, alkaline phosphatase, aspartate aminotransferase, total cholestrol, creatine kinase, γ-glutamyltransferase, glucose, HDL-C, LDL-C, potassium, sodium, trίglycerides, blood urea nitrogen and uric acid. We found that the Homo-aza-steroidal Ester, in contrast to the above antitumor drugs, only transiently affects the biochemical parameters associated with hepatic toxicity, though to a significantly lower extent than Chlorambucil, Methotrexate, Cisplatin or Epirubicin. lt thus appears that the proposed biochemical screening for eνaluating subacute toxicity during chemotherapy with antineoplastic drugs may offer an altenative approach for the early diagnosis of imminent side effects.
