ΤΗΕ DIAGNOSTIC SIGNIFICANCE OF IMMUNOPHENOTYPE INVESTIGAΤΙΟΝ ΙΝ ACUTΈ LEUΚEMIAS

Abstract

We have studied the diagnostic significance οf immunophenotyping acute leukemias where the FAB classification has been applied at the onset οf disease by using morphological and cytochemical criteria. The case population fοr this study consisted οf 30 patients with acute leukemia with mean age 54,7 ± 14,2. The cytochemichal study has been consisted οf Sudan Black Β (SBB) stain, non specific esterases (NSE) stain and periodic acil Schiff (PAS) reaction. For immunophenotyping we applied the procedure οf Alkaline-antialkaline Phosphatase (ΑΡΑΑΡ), by using monoclonal antibodies to detect the antigenic markers HLA-DR, CD10, CD19, CD20, CD2, CD5, CD7, CD13, CD14, CD33, CD41, Glycophorin-A and Terminal deoxynucleotide transferase (TdT). The monoclonal antibody against the myeloperoxidase (anti-MPO) has been used too. The FAB classilication in comparison with immunocytochemical investigation has showed the following interesting signs: 1) In seventeen οf thirty patients (57%) with acute leukemia there has been correlation between FAB classilication and the monoclonal antibodies sequence οf immunocytochemical reaction. 2) In seven pantients (29%) the immunophenotype investigation was significant for the termlnal diagnosis and treatment. 3) In six pantients (18%) the FAB classification has been modified in comparison with immunophenotype picture. The basic type οf leukemia (myeloid or lymphoid) did not change in these six cases. 4) The diagnosis and investigation οf blasts in biphenotypic and mixed leukemias is impossible without the aid οf immunophenotyping.
